Resistance against antiparasitic drugs is now an increasing problem. Despite a lot of effortd put on preventive measures, such as egg counting and selective treatments, there is still a need for alternative and sustainable methods that do not result in rapid development of drug resistance. The proposed project therefore aims to investigate vaccination as an alternative to antihelminthic treatment of the horse. Previous established methodology, within a research project on "immunopathological aspects of equine inflammatory bowel disease" will be useed and further developed in order to construct a vaccine against Strongylus vulgaris in particular. The applicability of a modern adjuvant "G3", in combination with irradiated larval antigen, will be evaluated and provide a basis for future vaccine trials in the horse. The adjuvant is well tolerated in the horse and can induce a cell-mediated immunity that is important for development of long-lasting and protective immunity.
Ökad resistens mot avmaskningsmedel är idag ett växande problem. Trots mycket fokus på förebyggande åtgärder, såsom träckprovstagning och selektiv avmaskning, finns det fortfarande ett stort behov av alternativa, hållbara metoder som inte leder till resistensutveckling. Det sökta projektet syftar därför till att undersöka vaccination mot inälvsparasiter som en profylaktisk åtgärd. Projektet ämnar att använda och utveckla tidigare etablerad metodik inom studier av "inflammationsimmunologi" i tarm från häst för att konstruera vaccin mot framförallt hästens stora blodmask. Användbarheten av ett modernt adjuvans, "G3", i kombination med parasitlarver som inaktiverats genom strålning, kommer att utvärderas och ge grund för framtida vaccinförsök i häst. Vi vet redan att G3-adjuvanset är väl tolererat i häst och preliminära resultat ger indikationer på att G3 kan inducera en cell-medierad immunitet vilket är väsentligt för att erhålla långvarig skyddande immunitet i mag / tarmkanalen.
